Images need to be "created" with IMAQ Create Image. You did one, called Video, which is the "Source" Image for the Extract Single Color Plane function, but you didn't wire in a second "Destination" image to this function. According to its Help, if the Destination is not wired, the output Image overwrites the Source.
So to fix this, create another Image using IMAQ Create, call it maybe "Processed", and wire it to the Dst Image terminal of Extract Single Color Plane. Let us know if this fixed your problem.
