I have, perhaps, a dumb question, but is NI USB-6216 compatible with Windows 10?

Yes, the USB-6216 is compatible. Windows 10 is supported by NI-DAQmx 15.1 and later, and the USB 6216 is supported by NI DAQmx 15.5 and later.
