What would the format of the command line be to open an excel file in a specific directory using the system.exe VI?

Pass cmd.exe (without quotes) as the command line and "full_path_to_xls_file\r\n" (with quotes and CRLF) as the standard input argument.  This will open any file, using the default application for that file extension.
