What is your RT target? Any of the Linux RT have FTP disabled by default for security reasons. NI recommends switching to WebDAV. But you should still be able to install the FTP driver on the RT system if you really want to stick with FTP.

Have you tried to use the absolute path /ni-rt/startup/.... rather than the relative one without leading slash?
The FTP servers current working directory may not be in the root when you attempt to list the contents. Alternatively try to execute the pwd command to see where you really are located!
