all M-Series products use a 68pin VHDCI (Very High density Micro Centronics)- Connector - have a look into the specifications manuals p. 9 (http://www.ni.com/pdf/manuals/371290d.pdf).
Thought, connectors and backshell kits should be available be several electronic suppliers, but be careful, when you design a cable. The SHC68-68-EPM (btw: EPM stands for Enhanced Performance M- Series) is especially designed for this boards in respect of impedance, shielding, twisting and proper connector resistance ....etc. to carry the signals properly. The PCI-6221 is also available with a 37-pin DSUB connector (https://www.ni.com/en-us/support/model.pci-6221.html). Making a cable for this connector may be easier.
