and also difference between read or write to a port and read or write to a line

A port is a group of 8 lines. For E-Series DAQ devices, they all have 8 DIO lines (or one port). When using Write to Port, you are righting an 8-bit value to all 8 lines. If you use Write to Line, you are only writing a 1 or 0 to one line only.
