I have used the DAQ express blocks in LabView to acquire sound using my computer's microphone and sound card, then performing frequency analysis on the acquired signal. Can I use the same method as an input into Signal Express 3.0?

You can use the DAQ Assistant in SignalExpress, but if you are acquiring sound card data it sounds like you may actually be using the Acquire Sound Express VI.
If you have a way to acquire the Sound through DAQmx then you should be able to do this in SignalExpress the same way you do in LabVIEW. If you are using the Acquire Sound Express VI, then you will have to configure the function in LabVIEW (you can then call it in SignalExpress). The Frequency Analysis functionality is built-in to the Full version of SignalExpress.
