I think the restrictions are also mentioned in the NI PXI/PCI-6561/6562 Specifications. There is a figure that shows the limitations for data delay and the "Waveform Specifications" section should provide the information you need regarding waveform quantum and sample size.

Hi,

When I setup inside a script to wait for a number of samples, does this wait also have to be a multiple of 4 samples. I am getting an error from this, but don't know if it is due to the number of samples I am waiting for not being a multiple of 4.

Thank you for contacting National Instruments.  Yes, the minimum waveform size is 4 and the driver enforces this by requiring a multiple of 4S for your wait time.
