Resolved, I had a TS handled opened in the LV GUI. If I close all LV GUI, then it's fine.

Another cause for this error "object reference not set to an instance of of an object" is if you have not logged into TestStand or have disabled Login On Start.
Go to Configure -> Station Options -> User Manager -> check box " Login On Start"
Close TestStand
Open TestStand and login
Update VI Calls will now work
