create ROI and match pattern

Hello All, 

I have used USB camera to adjust the frame and capture an Image, than I have logged (saved) the last image with the help of case structure.

Now, I want to identify and count the pattern precisely in the online show in VI. I have done it in Vision Assistant. But since my object's position is not fixed, I have to define ROI and Co-ordinate system. I am able to do it in Vision Assistant which works perfectly, but it is (ROI and Co-ordinate system) not working as I desired in VI Labview. 

Could anyone please help me.

could you please post all necessary files so it is possible to reproduce the issue (Vision Assisitant file, image file, template file)
