Image brighness adjustment from ROI (region of interest)

Is it possible to adjust brightness of an image with the average from a ROI (region of interest), and change the following images accordingly? My task is to detect blackspots in images, the problem is that the different images have different lighting so that the threshold for finding the blackspots varies from image to image. There is a total of 420 pictures that is going to be scanned. 

 

I have tried balancing the average light intensity in the images, but the result between the different images was too big so that the object detection threshold value were to different between them. Attached under you can find original images with processed images. I have also attached the balance VI. This vi is based on the example VI called "adaptive_image_thresholding" from the link under.
