Serial comm. broke after upgrading from version 4 (Win95) to 6.i (Win2000)

The cable is only a 3-Pin connection (RxD,TxD,GND). DTR,DSR and RTS,CTS are wired). May Win2000 have problems with connections and Win95 not?
Or occurs the error because of the change from a 16- to a 32bit system? The Port seems to be O.K., already tested with another device.

Did you upgrade LV4 to LV6i as well, or is this just an OS upgrade? If it is the later, and your application is a DOS app or does not use the Win32 API - then it will not work on Win2000. The NT architecture does not allow user applications to access the serial port directly.
